ABSTRACT

We compared the ways of deproteinization for crude polysaccharides of Coprinus comatus, and finally selected Sevage method as the optimal method. Two main fractions of Ccp-I-A and Ccp-I-B were obtained after DEAE-52 cellulose and Sephadex G-200 chromatography, both were white-floc, soluble in water, insoluble in absolute ethyl alcohol, acetone and other organic solvents. Additionally, Fehling reagent, CTAB, Sulphuric acid-carbazole, I-KI and FeCl₃ reaction were all negative. GC analysis showed Ccp-I-A was composed of mannitose, glucose and galactose in molar ratios of 2.03:9.52:1, whereas Ccp-I-B was composed of fucose and galactose with molar ratios of 1:5.21. Antioxidant activity test showed that Ccp-I-A and Ccp-I-B had good scavenging abilities on DPPH and ·OH. Compared to Ccp-I-B,the scavenging activity of Ccp-I-A was much stronger, and the scavenging rate could reach 72.1% and 55.3% respectively when the concentration was 300 μg/mL.

ABSTRACT

The principal objective of this study was to acquire basic data regarding the mycelial growth characteristics for the artificial cultivation of Coprinus comatus. 12 URP primers were employed to evaluate the genetic relationships of C. comatus, and the results were divided into three groups. Among six kinds of mushroom media, MYP medium was selected as the most favorable culture medium for C. comatus. The optimal temperature and pH ranges for the mycelial growth of C. comatus were 23~26degrees C and pH 6~8, respectively. The carbon and nitrogen sources for optimal mycelial growth were sucrose and tryptone, respectively.

Objective To observe the effect of extract of Coprinus comatus on small intestinal propulsion and gastric emptying function in mice.Methods Effect of extract of Coprinus comatus at different doses on small intestinal propulsion and gastric emptying function was observed in normal mice,neostigmine-load mice and epinephrine-load mice by charcoal powder method and by modified phenol red method respectively.Results The small intestinal propulsion of normal mice was promoted by the extract of Coprinus comatus at the doses of 3.0,6.0,9.0 and 12.0 g/kg,and the gastric emptying function of normal mice was promoted by the extract of Coprinus comatus at the doses of 1.5,3.0,6.0,9.0 and 12.0 g/kg,the difference being significant as compared with the normal group (P

ABSTRACT

Coprinus comatus cultivated on cotton seed hull medium decomposed lignocellulose straggly and was high of absolute biological efficiency. Lignocellulose is the main carbon source for the fruiting stage of the fungus. There existed the positive correlation between the degradation rates of the cellulose and hemicellulose in the medium and the activities of extracellular CMCase (carboxymenthelcel lulase), FPase (filter paper cellulase) and HCase (hemicellu-lase), there also existed the positive correlation between the degradation rate of the lignin in the medium and the activity of extracellular laccase, but no correlation between the degradatio rate of the lignin in the medium and the activity of peroxidase. The activity of extracellular amylase was comparatively high at mycelial growth stage, and the protease activity peek was at teh time when the fruitbody matured.

ABSTRACT

Suitable conditions for polysaccharide production in submerged culture by Coprinus comatus Ft. CC-8810 were: glucose 40g, Soybean cake powder 30g, yeast extract powder 5.0g, bran 30g, KH_2PO_4 1.0g, MgSO_4?7H_2O 0.5g, tap water 1L, pH5—6, 28℃, 160r/min. Seed culture grown in shake flask for 4 days were inoculated into 1.5L fermentor. After 5—6 days the fermentation ended up with decreases in pH to 4.9—5.0 and in reducing sugar to 1.2%. 45.1g(dry Wt.)/L hyphae and 3.6g/L polysaccharide were obtained.

ABSTRACT

Submerged fermentation parameters for the chromium-rich mycel ia of Coprinus comatus was studied in optimal medium. According to the yield of organic Cr, the optimal fermentation parameters were obtained as follows: the amount of CrCl_3 6H_2O in medium was 200mg/L; the inoculation volume was 8%~10%; the aeration ratio was 80:250 (medium volume: flask volume); the cul tural period was 5 days at 26℃ and at 100r/min. The highest yield of organic C r in mycelia of Coprinus comatus was 1597.88?g/100mL and the enrichment ra te of Cr was about 9.09%.
